摘  要:温汤孵化法是一种重要的家蚕卵人工孵化方法。酯酶A4 (EsteraseA4 ,EA4 )是家蚕卵的滞育生物钟蛋白质。将家蚕C10 8品种卵温汤处理 ,纯化出EA4 ,测定了EA4的ATP酶 (ATPase)活性变化。结果显示 :产卵后 15h(2 5℃ )的温汤处理蚕卵 ,其EA4的ATP酶最大活性 ,体外 5℃或 2 5℃条件下 ,分别出现在处理后 2 4h或 1.5h ,蚕卵也随即解除滞育 ,获得孵化能力 ;产卵后 30h(2 5℃ )的温汤处理蚕卵 ,其点青卵率随温汤刺激量增大而提高 ,点青时间随刺激量增大而缩短 ;蚕卵EA4的ATPase最大活性出现时间也随温汤刺激量增大而提前 ;且温汤处理蚕卵EA4的活性变化与蚕卵活化有高度相关性。The hot water treatment is an important method for artificial hatching in silkworm eggs. Esterase A4 (EA4), a time-interval measuring enzyme in the diapausing eggs of Bombyx mori, was purified from hot water-treated eggs of Bombyx C108 strain, and has been studied using ATP as a substrate. The ATPase activity peak for EA4 purified from 15 hours old eggs appeared at hour 24 at 5 ℃ or hour 1.5 at 25 ℃ after the treatment, and the diapause of eggs was terminated rapidly. ATPase activity peak for EA4 from 30 hours old eggs is appeared at an early time, the period of incubation shortens and the rate of head pigmentation of embryos enhances, by increasing the hot water stimulus (soaking duration). The changes of ATPase activity for EA4 in hot water treated eggs are highly correlated with eggs activation.
