摘  要:考察了甾体11b-羟基化过程中,底物17a,21-二羟基孕甾-4-烯-3,20-二酮-21-醋酸酯(2)投料时间、投料量及pH对细胞色素P-450(3)诱导和氢化可的松(1)产量的影响。在此基础上提出了生产1的诱导发酵改进方法:菌体生长至16h投入2 0.3g/L,培养至24h补加2 0.7g/L,培养至32h调至pH6.5,以后每12h调节一次。结果表明,在发酵周期不变(68h)的情况下,3蛋白量和1产量分别比一次投料法提高了11.2%和12.7%。The feeding time and concentration of 17a,21-dihydroxypregn-4-ene-3,20-dione-21-acetate(2)and thepH value of the environment which affected the induction of cytochrome P-450 and the yield of hydrocortisone during the11a-hydroxylation of steroids were investigated. The optimal fermentation conditions for induction were as follows: 0.3g/Land 0.7g/L 2 were fed at 16h and 24h after inoculation, respectively; adjusted the pH value to 6.5 at 32h and then regulatedto 6.5 every 12h. It was showed that the content of cytochrome P-450 protein was increased by 11.2% and the yield ofhydrocortisone was increased by 12.7% over the single feeding method.
