摘　　要：目的:研究真皮多能干细胞的部分基本生物学特性,观察其在创伤愈合中的意义。方法:采用前期建立的技术分离、纯化并扩增真皮多能干细胞;利用流式细胞技术观察真皮多能干细胞周期变化;四唑盐比色试验(MTT法)观察其辐射敏感性。收集伤后1,2,3和4d伤口液,观察其对真皮多能干细胞的反应性。结果:分离、纯化的真皮多能干细胞形态较为均一,增殖能力强;培养的细胞凋亡数量少,多处于静止期,对辐射极为敏感;这种细胞增殖对伤口液具有良好的反应性,尤其伤后早期伤口液作用更为明显。结论:发育成熟的个体中仍存在较为幼稚的真皮多能干细胞,它们在创伤愈合过程中具有重要意义,特别是在创伤修复启动中的作用值得进一步深入研究。AIM:To study the elementary biological characteristics of dermal pluripotent s tem cells,and to investigate the significance of these cells in the processes of wound healing. METHODS:Dermal pluripotent stem cells were routinely isolated,purified and exp anded by the technique established in the prophase.The cell cycles were determin ed with flow cytometry, and the radiosensitivity was dertermined by MTT assay.Wo und fluid was collected on the 1st,2nd,3rd and 4th day,and the responses of derm al pluripotent stem cells to wound fluid were investigated. RESULTS:The isolated and purified dermal pluripotent stem cells had similar sh ape,and their abilities of proliferation were very strong.Most of the cultured c ells were quiescent,and were sensitive to radiation. Furthermore,the cells had good response to wound fluid,especially to the early-phase wound fluid. CONCLUSION:There are primordial dermal pluripotent stem cells in mature specie s.Dermal pluripotent stem cells play important roles in the process of wound hea ling,and especially their significances in the initiation of wound repair are wo rth investigating further.
