摘 要:目的 探讨内毒素和肿瘤坏死因子α(TNFα)对大肠杆菌和白色念珠菌混合感染小鼠病情的影响。方法 建立小鼠大肠杆菌、白色念珠菌单独及混合感染模型 ,对比多粘菌素B拮抗内毒素活性 ,观察各组小鼠病死率 ,血浆内毒素水平 ,以逆转录多聚酶链反应 (RT PCR)技术检测各组小鼠肺组织中TNFα水平。结果 大肠杆菌和白色念珠菌混合感染小鼠病死率明显高于各自单独感染组 ,大肠杆菌和白色念珠菌混合感染 2 4h后血浆内毒素水平明显高于大肠杆菌单独感染 ,大肠杆菌和白色念珠菌混合感染 7、10h后肺组织中TNFαmRNA水平明显高于各自单独感染。结论 内毒素和TNFα可导致大肠杆菌和白色念珠菌混合感染小鼠病情加重 。Objective To study the effect s of endotoxin on mice with concurrent infection of escherichia coli and candida a lbicans.Methods Animal models with concurrent infection of esch erichia coli and candida albicans and with either organism infection alone were established.Polymyxin B was used to neutralize the toxic and immunologic effects of endotoxin.Then the death rates,plasma endotoxin levels among different group s were compared.The TNFα mRNA levels in lungs among different groups were also compared by RT PCR.Results Mortality of mice with conbined inf ection of escherichia coli and candida albicans was significantly higher than th at of mice with infection alone.Plasma endotoxin levels of mice with combined in fections of escherichia coli and candida albincans were significantly higher tha n that with escherichia coli infection alone after 24 hours infection.TNFα mRNA levels in lungs with combined infection of the two organisms were significantly higher than that with organism infection alone after 7 and 10 hours infection. Conclusion Endotoxin and TNFα can exacerate concurrent infecti ons of escherichia coli and candida albicans and raise the mortality of infected animals.
