肾络通对阿霉素肾病大鼠肾小管间质增殖细胞核抗原表达的影响

摘　　要：目的 :观察中药肾络通对单侧肾切除加重复阿霉素注射所诱导的肾小管间质纤维化大鼠增殖细胞核抗原 (PCNA)和转化生长因子 β1(TGF β1)的影响。方法 :实验动物分为病理模型组、中药治疗组、西药治疗组和假手术组 ,采用生化、放免、免疫组化、病理图像分析等方法 ,检测肾功能、肾小管间质PCNA、TGF β1的蛋白表达。结果 :中药肾络通组 2 4h尿蛋白定量、血肌酐、尿 β2 MG、尿NAG明显低于病理模型组 ,部分优于西药组。结论 :肾络通可能是通过抑制PCNA和TGF β1表达 ,保护肾功能 ,延缓肾小管间质纤维化的进展。Objective:to observe the effect of Shenluotong on the expression of proliferating cell nuclear antigen(PCNA)and transforming growth factor-β 1(TGF-β 1)in one-side kidney resection and adriamycin-induced tubulointerstitial fabrosis in rats.Methods:The rats were randomly divided into four groups,pathological model,Shenluotong treatment,Valsartan treatment and sham operation.Biochemistry,radio-immunity,immunohistochemistry and analysis of the pathological picture were adopted to examine the renal function and the protein expression of PCNA and TGF-β 1.Results:24 hours' urine protein,serum creatinine,urine β 2-MG,urine NAG of the Shenluotong group were lower than that of the pathological model significantly,and some of them were even better than that of Valsartan treatment.Conclusions:Shenluotong may protect renal function and inhibit tubulointerstitial fabrosis through decreasing the expression of PCNA and TGF-β 1.
