摘 要:目的 研究氯胺酮对沙土鼠脑缺血 -再灌流损伤后细胞凋亡状态及相关基因caspase - 3、c-fos表达的影响。方法 沙土鼠随机分为假手术组、缺血 -再灌流组和氯胺酮组 ,各 10例。缺血 -再灌流组 :分离双侧颈总动脉 ,用无创动脉夹阻断脑血流 10min后松开动脉夹恢复血流。假手术组 :仅分离双侧颈总动脉但不予阻断。氯胺酮组 :血流阻断前 30min腹腔内注射氯胺酮 10mg/kg,余同缺血 再灌流组。采用缺口末端标记法 (TUNEL)原位标记DNA片段检测凋亡细胞 ,免疫组化SP法进行caspase - 3、c -fos的免疫组织化学染色。结果 缺血 -再灌流组及氯胺酮组脑组织中TUNEL及caspase- 3、c-fos阳性细胞数明显多于假手术组 (P <0 0 1) ;氯胺酮组 3项指标阳性细胞数目均明显低于缺血 -再灌流组 (P <0 0 1)。结论 氯胺酮减少沙土鼠脑缺血 -再灌流损伤后细胞的凋亡 ,其对caspase - 3激活和cObjective To investigate the effect of ketamine on neurons apoptosis and expression of caspase-3 and c-fos after cerebral ischemia/reperfusion in gerbils.Methods Thirty gerbils were randomly assigned to three groups:sham operated group(SO,n=10),ischemia/reperfusion group(IR,n=10),and ketamine treatment group(K,n=10).The bilateral common carotid arteries of the gerbils were isolated.In group IR,the bilateral common carotid arteries were occluded with aneurysm clips for 10min.The clips were released to resume global blood flow.In group K,ketamine(10mg/kg) was injected intrapenitoneally 30 minutes before arteries were occluded.And in group SO,gerbils were operated without occluding the arteries.All the gerbils were anesthetized with sodium pentobarbital,and the brain were removed 24h after operation.Apoptosis were detected by terminal-deoxynucleotidyl transferase mediated nick end labeling(TUNEL),and caspase-3 and c-fos were detected by immunohistochemistry.Results Apoptotic neurons and the expressions of caspase-3 and c-fos in groups IR and K increased significantly,compared with those in group SO( P <0 01).Apoptotic neurons and the expression of caspase-3 and c-fos in group K were significantly decreased than those in group IR( P <0 01).Conclusions Ketamine decreased neuron apoptosis after cerebral ischemia/reperfusion injury.This may be related to the suppressed expressions of caspase-3 and c-fos by ketamine.
