摘 要:目的 :比较细胞因子PF4与辐射防护剂WR 2 72 1对全身照射 (TBI)后小鼠造血系统的保护作用 .方法 :雄性BALB/c小鼠随机分为正常对照组、照射对照组、WR 2 72 1保护组和PF4保护组 ,后 3组给予 7Gy60Coγ 射线全身照射 .用流式细胞仪 (FCM )测定细胞周期和细胞凋亡 ,采用内源性脾结节法测定脾集落形成单位 (CFU S) ,进行小鼠骨髓病理检查 .结果 :WR 2 72 1和PF4可以减少造血细胞坏死、凋亡 ,增多CFU S ,还能减轻骨髓基质水肿、出血 ;PF4造血保护作用更强 .结论 :PF4和WR 2 72 1在小鼠全身照射前使用 ,可以减轻骨髓辐射损伤 ,可作为肿瘤患者放射治疗时的骨髓保护剂 .PF4对于全身照射 (TBI)AIM: To compare the hematopoietic protective effect of WR 2721 and PF4 from total body irradiation in mice. METHODS: BALB/c male mice were randomly divided into control group, exposure group, protection group with WR 2721 and protection group with PF4. The last three groups were irradiated with 7 Gy 60 Co γ rays. The change of cell cycle and cellular necrosis and apoptosis were determined by flow cytometry. The CFU S was counted by the technique of endogenous splenic nod. The bone marrow histopathological observation was also carried out. RESULTS : WR 2721 and PF4 decreased the hematopoietic cellular necrosis and apoptosis, improved the forming ability of CFU S, and decreased edema and bleed of bone marrow. The effect of PF4 was stronger than WR 2721. CONCLUSION : Application of WR 2721 and PF4 before total body irradiation in mice, could decrease the radiation damage of the bone marrow, which would be protective agents of the bone marrow while radiotherapy. The hematopoietic protective effect of PF4 is better than that of WR 2721.
