摘 要:全胚胎培养方法是八十年代才应用的一种新的体外实验方法,具有省时、省力、省钱的优点。本研究探讨垒胚胎培养方法可否用于检测铅的胚胎毒性。实验结果表明,在培养基中加铅(铅含量为30.68,62.88,88.64μg/ml)对胚胎体外染毒及给孕鼠腹腔注射醋酸铅溶液(含铅31.81mg/kg)染毒。铅均可影响胚胎卵黄囊发育,并导致胚胎生长发育迟缓及出现畸形,且有剂量反应关系。与整体动物实验相同,全胚胎培养方法可以用于检测铅的胚胎毒性。The present study attempts to test the feasibility of using whole rat embryo culture to evaluate the embryotoxicity of lead. There are two parts in this thesis: Part A is that 9.5 day rat embryos were cultured for 48 hours in rat serum to which lead was directly added, the final concentrations were 0, 30.68, 62.88, 88.64ug/ ml, respectively. Part B is that pregnant rats received an intraperitoneal injection of lead acetate (31.8mg pb/ kg) on the 9.5th day of gestation. After 24 hours, the rats were sacrificed and the embryos were explanted into the rat serum taken from untreated animals to culture for 24 hours. The results showed that lead damaged visceral yolk sac, inhibited the development of the embryos, and induced malformation of embryos. The embryo toxic and teratogenic effects of the lead on the cultured embryos were similar to those on the embryos exposed to lead in vivo.
