摘  要:目的 观察吖啶橙 (AO)与长波紫外线 (UVA)消毒血浆中f2 噬菌体后形态的变化。方法 以f2 噬菌体为试验病毒 ,通过AO与UVA单用及联用灭活病毒效果测定筛选二者协同作用的最佳条件 ,在此最佳消毒条件下利用透射电镜观察f2 噬菌体在消毒前后形态的变化。结果 经AO处理后再经UVA照射 ,能明显减少血浆中f2 噬菌体的滴度 ,两因素的T E值大于 1;经有效灭活病毒的剂量 ( 6μg mlAO +4 5 0 0J m2 UVA)消毒后与消毒前相比 ,血浆中大多数f2 噬菌体由球形变为纺锤状和丝状 ,且轮廓变得不清晰 ,电子密度降低。结论 AO与UVA两因素具有协同增效作用 ;消毒后f2 噬菌体的形态明显发生了变化 ,由球形变为纺锤状和丝状。Objective To observe the morphological changes of bacteriophage f 2 in the plasma after inactivation by acridine orange (AO) and long wave ultraviolet rays (UVA). Methods Bacteriophage f 2 was used as the test virus. The inactivation effects of AO, UVA, and combined use of AO and UVA were detected for the purpose of choosing the optimal condition of inactivating bacteriophage f 2 in plasma. After treatment with AO and UVA under this condition, the morphological changes of bacteriophage f 2 in plasma were observed by transmission electron microscopy. Results The titers of bacteriophage f 2 in plasma were significantly reduced after treatment with AO followed by UVA irradiation. The value of T/E of these two agents was higher than 1. Transmission electron microscopy showed that the effective dosage for inactivating the virus was 6 μg/ml AO+4 500 J/m 2 UVA. After inactivation of the virus, changes in shape were found in the majority of bacteriophage f 2 in plasma, from spheriform shape into spindle and silk like ones, with unclear contour and decreased electronic density. Conclusion UVA and AO have synergistic effect on the inactivation of bacteriophage f 2. Treatment with AO and UVA may result in changes in the shape of bacteriophage f 2, from spheriform shape into spindle and silk like ones.
