摘 要:目的 研究热休克蛋白 70 (HSP70 )在大鼠烫伤后早期肠粘膜损害中的作用。方法 90只Wistar大鼠随机分为单纯烫伤组 (B组 )、亚砷酸钠预处理组 (SA组 )及亚砷酸钠 +槲皮酮预处理组 (SA +Qu组 ) ,另取 6只大鼠作为正常对照。Westernblot检测HSP70蛋白表达情况 ,于 3 0 %TBSAⅢ度烫伤后 3、6、12、2 4、48h检测血浆内毒素及D 乳酸含量变化 ,肠粘膜组织石蜡切片HE染色后观察肠粘膜损害情况。结果 亚砷酸钠预处理可显著诱导HSP70蛋白表达 ,槲皮酮预处理可明显抑制HSP70的诱导 ;SA组伤后各时相点小肠组织病理评分明显低于B组 ,大鼠烫伤后血浆内毒素及D 乳酸水平均有不同程度的升高 ,SA组伤后多数时相点血浆内毒素及D 乳酸水平显著低于B组 ,SA +Qu组小肠组织病理评分及血浆D 乳酸和内毒素水平与B组无显著差别。结论 亚砷酸钠诱导的HSP70表达增加可明显减轻大鼠严重烫伤后早期的肠粘膜屏障破坏 ,降低肠粘膜通透性 。Objective To study the effects of heat shock protein 70 (HSP70) on intestinal mucosa injury in scalded rats. Methods A total of 90 Wistar rats were randomly divided into 3 groups: simple burn group (Group B), sodium arsenite pretreatment group (Group SA) and sodium arsenite plus quercetin pretreatment group (Group SA+Qu). The other 6 Wistar rats were used as the normal control. HSP70 expression in small intestine samples was determined by Western blotting. Plasma endotoxin and D lactic acid content were determined at 3, 6, 12, 24 and 48 h after 30% TBSA Ⅲ degree scald. Damage to intestinal mucosa was observed by pathological section with HE staining. Results Sodium arsenite pretreatment could significantly increase HSP70 expression. Quercetin could inhibit HSP70 expression induced by sodium arsenite in the small intestine. Intestinal pathological grading in Group SA was significantly lower than that in Group B at different time points after scald. Plasma endotoxin and D lactic acid content in Group SA were significantly lower than those in Group B at most time points after scald. There was no difference between Group B and Group SA+Qu. Conclusion The enhanced HSP70 expression induced by sodium arsenite can attenuate the intestinal mucosal barrier injury and decrease the permeability of intestinal mucosa and the translocation of endotoxin from gut in scalded rats.
