中药露蜂房醇提取物对人红白血病K562细胞的生长抑制及凋亡诱导作用

摘　　要：目的 :研究中药露蜂房 (NidusVespae,NV)醇提取物对K5 62细胞的作用及其机制。方法 :利用3H TdR掺入、透射电镜、原位末端标记 (TUNEL)技术、免疫组织化学和图象分析等方法。结果 :不同浓度NV醇提取物对K5 62细胞生长具有明显抑制作用 ( 2 7%～ 5 6% )。NV醇提取物组K5 62细胞呈典型的凋亡形态学改变 ,其Bcl 2蛋白表达显著减弱、Bax蛋白表达显著增强。结论 :中药NV醇提取物可明显抑制K5 62细胞增殖 ,其作用机制可能是通过Bcl 2、Bax的表达 ,从而诱导白血病细胞凋亡。Objective:To study the inhibition effect of the Nidus Vespae alcohol extract on K562 cell. Methods:The inhibition effect of the Nidus Vespae alcohol extract on K562 cells was studied with 3 H -TdR incorporation technique;the morphological changes was studied with transmission electronic microscope and TUNEL. The expression of Bcl-2 and Bax were studied with immunohistochemical method and image analysis system. Results:Different concentration of the Nidus Vespae alcohol extract inhibited the proliferation of K562 cells, and the inhibition rate was 27%-56%. Leukemic cells in experimental group showed typically apoptotic morphological change. Comparing with blank control group, the number of positive cells increased and the expression of Bcl-2 declined, while the expression of Bax increased. Conclusion:The Nidus Vespae alcohol extract can significantly inhibit the proliferation and induce the apoptosis of K562 cells, and regulate the apoptosis by inhibiting the expression of Bcl-2 and promoting the expression of Bax.
