摘  要:目的 了解更年期大鼠雌激素水平与脾细胞凋亡的增龄性变化。方法 应用放射免疫法测定血清雌二醇 (E2 )水平 ;AnnexinV FITC/PI双染色流式细胞仪测定脾细胞凋亡率 ;逆转录聚合酶链反应 (RT PCR)检测脾细胞凋亡基因Bcl 2、Bax的表达。结果 更年组大鼠血E2 水平低于青年组 (14 .89pg/mLvs 34.0 9pg/mL ,P <0 .0 1) ;脾细胞凋亡率高于青年组 (7.7%vs 10 .3% ,P <0 .0 5 ) ,促凋亡基因Bax的转录水平也高于青年组。结论 衰老过程中脾淋巴细胞凋亡增加 ,受到相关基因的调控 。Purpose: To investigate the relation between the changes of plasma estradiol levels and apoptosis in splenocyte of menopausal rats with aging. Methods: Serum estradiol levels were measured by radio- immunoassay(RIA). The apoptosis in splenocyte of rats was measured by Annexin V-FITC/PI flow cytometric assay, Bcl-2 and Bax genes expressions were detected by RT-PCR technique. Results: The serum E2 level of menopausal group was lower than that of young group. The percentages of apoptosis cells in splenocyte of menopausal rats were significantly higher than those of the young rats. The expression of Bax genes was higher than that of the young rats. Conclusions: It suggests that the increased rate of apoptosis in splenocyte of menopausal rats was related to the decreased estradiol blood levels with aging.
