摘  要:目的 研究日本血吸虫 (中国大陆株 )重组信号蛋白 14 - 3- 3(rSj14 - 3- 3)DNA作为血吸虫病疫苗候选分子的潜能。方法 碱裂解法大量制备rSj14 - 3- 3和rSjGSTDNA ,将两种DNA分别免疫BALB/c小鼠进行攻击感染实验。 结果 在尾蚴攻击感染 6w后 ,剖杀小鼠计算各组的减虫率 ,rSj14 - 3- 3DNA组为 34 2 0 % ,rSj14 - 3- 3+rSjGSTDNA组为32 4 0 % ;减卵率为 (按以上组序 ) 5 0 74 %和 5 5 2 3%。结论 重组Sj14 - 3- 3DNA具有一定的抗血吸虫病潜能 ,有可能作为日本血吸虫病的候选疫苗 ,但未见Sj14 - 3- 3、SjGST两种重组DNA的协同作用。To evaluate protective effects of the recombinant signal protein(14-3-3 rSj14-3-3) DNA of Schistosoma japonicum(Chinese strain) and to observe the cooperative effect of rSj14-3-3 and rSjGST DNA as candidate for vaccine against Schistosoma japonicum,the Sj14-3-3 and SjGST recombinant plasmids were prepared by alkali burst method.These DNA were immunized into BALB/c mice and followed by challenging infection.After 6 weeks of challenging infection,the immunized mice were killed,and the worm and egg reduction rates were calculated It was found that the worm reduction rate in Sj14-3-3 DNA immunized group was 34.2%,and that in Sj14-3-3 DNA+SjGST DNA group was 32.4%;the egg reduction rates were 50.74% and 55.23% respectively.It concludes that recombinant rSj14-3-3 DNA has certain potentials to induce the anti-schistosomal effect in mice,and might be used as the candidate of vaccine,but there is no any cooperative effect with rSjGST.
