摘　　要：目的　为了观察VacA+ BCS对上皮细胞骨架的作用 ,并分析这种致细胞骨架病变的作用特点 ,从而明确VacA等分泌性毒力因子的致病意义。方法　利用cDNA基因表达谱芯片技术研究VacA+ BCS作用细胞中细胞骨架蛋白相关基因的表达变化 ,同时利用免疫荧光染色以及共聚焦显微镜观察细胞骨架的形态结构变化。结果　同VacA-BCS作用细胞和空白对照相比 ,VacA+ BCS作用细胞引起了细胞骨架蛋白相关基因表达的改变 ,同时 ,显微镜观察结果也从形态和结构方面证实了上皮细胞骨架的改变。结论　VacA+ BCS可以引起上皮细胞骨架的结构和功能的改变 ,VacA等细菌分泌性毒力因子可能在H pylori致病中发挥重要作用。Aim To elucidate the morphologic and structural changes of cytoskeleton of gastric epithelial cells and analyze the characteristics of these changes.Methods Cells were incubated with VacA -BCS and VacA -BCS of Helicobater pylori respectively.The alteration of cytoskeleton associated gene profile was evaluated by cDNA microarray.The morphologic and structural changes of the cytoskeleton were observed by immunofluorescence staining and confocal microscopy.Results As compare with those treated with VacA -BCS,cells treated with VacA +BCS showed significant alterations in the expression of cytoskeleton associated genes.Such alteration was further confirmed by the morphologic and structural changes of cells.Conclusion VacA +BCS can induce morphologic and structural changes of treated cells.VacA and other secretary factors may play important roles in the pathogenicity of Helicobater pylori associated diseases.
