胰脾联合移植后供、受体脾脏的细胞凋亡变化

摘　　要：目的　了解胰脾联合移植后供、受体脾脏的免疫反应和细胞凋亡变化。方法　对封闭群大鼠行胰脾联合移植 ,于术后 3、7、12、2 0d摘取供脾和宿主脾脏 ,观察组织学和细胞凋亡变化 ,并与正常脾脏比较。结果　术后供脾肿大明显 ,宿主脾脏不肿大。供、受体脾脏中央动脉周围淋巴鞘均明显增大 ,宿主脾脏淋巴母细胞转化活跃 ,供脾淋巴母细胞比例较少 ,2 0d时 ,供脾白髓纤维化 ,宿主脾脏则无。供脾细胞凋亡在术后第 3、7、12、2 0天分别为 ( 4 0 .95± 9.40 )、( 4 5 .19±7.93 )、( 3 9.0 9± 8.12 )和 ( 2 8.77± 5 .97)个 /高倍视野 ;宿主脾脏细胞凋亡分别为 ( 4 0 .49± 7.2 6)、( 68.91± 6.94)、( 2 8.40± 5 .2 8)和 ( 2 4.5 6± 3 .5 2 )个 /高倍视野 ;均显著高于正常脾脏 (P <0 .0 0 1)。结论　胰脾联合移植后 ,供、受体脾脏内细胞凋亡活性均明显升高。Objective To observe the cell apoptoticactivity in donor and recipient spleens after combined pancreas spleen transplantation.Methods Non inbred Wistar rats were used as both donors and recipients.Before transplantation,the recipient animals were induced diabetic mellitus with streptozotocin.Combined pancreas spleen transplantation was performed.At 3,7,12,and 20 days after transplant,the animals were sacrificed respectively and both donor and recipient spleens were removed for histological examination and apoptotic determination (TUNEL assay).The spleens from normal animals were used as control.Results From 3 to 20 days postoperation,the donor spleens showed markedly enlarged.The peri artery lymphocyte sheathes of donor and recipient spleens showed also enlarged under a light microscope.In recipient spleen,lymphoblastic transformation was active.Otherwise,in donor spleen,a decreased ratio of lymphoblast was found.At 20 days postoperation,the white pulp in donor spleen displayed fibrous,but the recipient spleen didn't.In both donor and recipient spleens,the activity of cell apoptosis was significantly increased as compared with control group ( P <0.001).Conclusion After combined pancreas spleen transplantation,the activities of cell apoptosis both in donor and recipient spleens are increased.
