摘　　要：目的　探讨B7单抗阻断B7/CD2 8通路在体外异种细胞免疫反应中的作用。方法　分离豚鼠 ,大鼠外周血单个核细胞 (PBMC) ,豚鼠细胞经丝裂霉素处理作为刺激细胞 ,大鼠细胞作为反应细胞 ,通过异种单向混合淋巴细胞培养 (Xeno MLC) ,3 H TdR掺入 ,酶联免疫吸附分析方法 (ELISA) ,流式细胞仪 (FCM )检测对照组 ,反应组 ,B7单抗组的淋巴细胞增殖效应 (cpm) ,以及IFN γ ,IL 4和CD3+ ,CD3+ CD2 5 + ,CD4 + ,CD8+ ,CD4 + /CD8+ 的动态变化。结果　B7单抗组的淋巴细胞增殖效应 (cpm) ,以及IFN γ ,IL 4和CD3+ ,CD3+ CD2 5 + ,CD4 + ,CD8+ 的变化均显著低于反应组 ,但其对CD4 + /CD8+ 比值无影响。结论　B7单抗可以部分抑制异种细胞间免疫反应 。Objective To study the role of B7 mAb in blocking the pathway of B7/CD28 in Xeno-cells immune response in vitro.Methods Peripheral blood mononuclear cells(PBMC)of guinea-pig and SD rat were isolated respectively.Then the mixed lymphocyte culture was performed.The PBMCs of guinea-pig treated with mitomycin acted as stimulating cells,the PBMCs of SD rat,as responding cells.The cell proliferation,secretion of IFN-γ(ng/L) and IL-4(IU/ml) were measured by t 3H-TdR incorporation and enzyme linked immunosorbent assay(ELISA).CD3,CD3 +CD25 +,CD4 + and CD8 + cells were analyzed with flow cytometry.Results After 5 days culture,the cell proliferation,secretion of IFN-γ,IL-4 and the number of CD3 +,CD3 +CD25 +,CD4 +,CD8 + of SD rat with PBMCs treated with B7 mAb were significantly lower than the cells without treating with B7 mAb(-P-≤0.05).Conclusion B7 mAb can inhibit the activation of T cells in Xeno-cell immune response,but the mechanism needs to be studied further.
