摘  要:目的 对几种重要的生长因子在正常人体皮肤、肌腱中的表达差异进行研究。方法体外培养人肌腱细胞和人皮肤成纤维细胞,观察细胞的形态特点;用基因芯片、免疫组化、免疫细胞化学、RT-PCR的方法对肌腱细胞和成纤维细胞的几种生长因子进行检测及比较。结果所测生长因子中除转化生长因子-βⅡ型受体(TGF-βRⅡ)、类胰岛素生长因子-结合蛋白3(IGF-BP3)外,其余在肌腱细胞和成纤维细胞中的表达均无明显的差异。结论 人成纤维细胞与人肌腱细胞的生长因子表达基本相似,通过对皮肤成纤维细胞的某些特性的改造,可以使其成为构建组织工程化肌腱的种子细胞。Objective To study the differential expression of several growth factors between human tenocytes and dermal fibroblasts. Methods Human tenocytes and dermal fibroblasts were cultured in vitro and their morphological characteristics were observed. Comparison was made between the two types of cells using gene chip, immunohisto-chemical staining and RT-PCR. Results Most of the growth factors that were invesgated were similarly expressed in tenocyte and fibroblast except TGF-βR Ⅱ and IGF-BP3. Conclusion The growth factor profile is similar between tenocytes and dermal fibroblasts. Thus, dermal fibroblasts may potentially be used in tendon engineering as seed cells.
