摘 要:[目的 ]研究乙醇对大鼠脑组织神经递质的影响。 [方法 ]雄性SD大鼠 2 5只 ,随机分为低剂量组、高剂量组和对照组 (分别为 8、8、9只 )。用相应 2 5 .6% (V/V)、5 1.3 % (V/V)乙醇浓度和对照组用蒸馏水 ,一次灌胃 (灌胃体积为1 0ml/ 10 0g体重 )染毒。 1h后处死大鼠 ,取血、脑纹状体和海马 ,分别测定血中乙醇浓度、脑纹状体单胺类递质含量及海马中强啡肽A水平。 [结果 ]纹状体多巴胺 (DA)含量随乙醇染毒剂量增加而升高 ,呈剂量 效应关系。低剂量组 3 ,4 双羟苯乙酸 (DOPAC)、强啡肽A(dynorphinA ,DynA)含量显著升高 ;高剂量组 5 羟色胺 ( 5 HT)含量显著高于低剂量组和对照组。 5 HT、5 羟吲哚乙酸 ( 5 HIAA)含量与血中乙醇浓度 (BAC)显著相关。 [结论 ]脑组织神经递质的变化 ,可能是乙醇神经毒性的机制之一。To study the possible neurotransmitters mechanism of the alcohol-induced neurotoxicity. 25 male Sprague-Dauley rats(weighs about 160g) were randomly divided into 3 groups. Each group was singly administered intragastrically with alcohol solution (0%,25.64%,or 51.28% V/V). The intragastrical volume was 1.0 ml/100 g. Rats were sacrificed by decapitation in one hour after alcohol administration. The blood was sampled and cerebral cortex,striatum and hippocampus were rapidly removed and frozen immediately on dry-ice. The following indicators were assayed: 1)blood alcohol concentration(BAC); 2)monoamine neurotransmitters and its metabolites levels in striatum; 3) dynorphin A levels in hippocampus. The BAC of the high dose group,low dose group and control group were 67.50mM,63.15mM and 2.88mM,respectively. The level of BAC in the exposed groups were statistically higher than that in the control group.There was a increasing trend of striatal DA with the increase in exposure to ethanol dose. The DOPAC contents increased in low dose group. The level of 5-HT in striatum in high dose group was higher than that in low dose and control group.The dynorphin A levels in hippocampus in low dose group increased as compared with control group. [Conclusion] These results suggested that alcohol may affect the metabolism of monoamine neurotransmitters,which might lead to neurotoxicity of alcohol-exposed rats.
