摘 要:作者根据国内现有条件,建立一种简便易行的鸟苷酸环化酶(GC)活性测定方法。其原理是以非标记三磷酸鸟苷(GTP)为底物,经提取的鸟苷酸环化酶作用,生成的环鸟苷-磷酸(cGMP),用^(125)I-cGMP放射免疫分析法(RIA)测定,以cGMP的产率推算鸟苷酸环化酶活性。此法简便、灵敏、特异性和重复性好。The authors based upon dometic experimental condition, to describe a simple method for the measurement of guanylate cyclase activity. By taking the unlabled GTP as a substrate, under the action of extracted guanylate cyclase from mice liver, cGMP was formed and measured by 125I-cGMP RIA. Activitic guanylate cyclase is indirectly assaied. The method is easily, sensitively, specific, repeatedly and accuratly used for assay of guanylate cyclase activity.
