摘  要:目的 观察重组人内毒素结合肽蛋白在烧伤合并内毒素血症大鼠中的作用。方法 采用烧伤合并内毒素血症大鼠模型 ,分为模型组 (n =3 6)、治疗组 (n =3 6)及对照组 (n =6)。模型组在烧伤后立即腹腔内注射 1mg/kg内毒素(用 1ml生理盐水配制 ) ;治疗组腹腔内注射 1mg/kg内毒素 (用 0 5ml生理盐水配制 )后 ,再注入 40 0 μg/kg制备好的内毒素结合肽 (用 0 5ml生理盐水配制 ) ;模型组和治疗组分别在处理后 1、3、6、12、2 4、48h取血和肝、肺组织 ,对照组仅在脱毛后腹腔内注入 1ml生理盐水 ,取血和肝、肺组织作总体对照。采用生化速率法、ELISA法及组织学切片染色和电镜制样等方法 ,测定血清中丙氨酸转氨酶 (ALT)、肿瘤坏死因子 α(TNF α)、白介素 6(IL 6)的含量变化以及光、电镜观察肝、肺组织的病理学变化。结果 模型组于伤后 1h血清TNFα显著高于对照组 (P <0 0 1) ,6h达峰值 ;治疗组与模型组比较各时相点血清TNFα含量均显著降低 (P <0 0 1,P <0 0 5 )。血清IL 6含量测定结果 ,模型组于伤后 1h血清IL 6显著高于对照组(P <0 0 1) ,12h达峰值。治疗组 3~ 48h各时相点血清IL 6含量均显著低于模型组 (P <0 0 1,P <0 0 5 )。大鼠血清中谷丙转氨酶的动态变化提示 。Objective To observe the protective effect of recombinant human endotoxin binding peptide (EBP) on a rat model of burn combined with endotoxemia. Methods A total of 78 rats of model of burn combined with endotoxemia were divided into three groups: model group ( n =36), treatment group ( n =36), and control group ( n =6). Rats in the model group were intraperitoneally injected with 1 mg/kg endotoxin (prepared with 1 ml saline) immediately after burn. After intraperitoneal injection of 1 mg/kg endotoxin (prepared with 0.5 ml saline), rats in the treatment group were intraperitoneally injected with EBP (prepared with 0.5 ml saline). Blood and liver and lung tissues of rats in the model and treatment groups were collected at 1, 3, 6, 12, 24, and 48 h after treatment. Rats in the control group were intraperitoneally injected with 1 ml saline after trichomadesis. Blood and liver and lung tissues of rats in the control group were collected for the total control. The changes of alanine aminotransferase (ALT), TNF α, and IL 6 contents in serum were determined by biochemical velocity analysis, ELISA, and light microscopy. The pathological changes of the liver and lung tissues were observed light microscopically and electron microscopically. Results The serum TNF α level of rats in the model group was significantly higher than that in the control at 1 h after injury ( P <0.01) and reached the peak value at 6 h. Serum TNF α level of rats at each time point in the treatment group was significantly lower than that in the model group ( P <0.01, P <0.05). Serum Il 6 level of rats in the model group was significantly higher than that in the control group at 1 h after injury ( P <0.01), and reached the peak at 12 h. The serum IL 6 levels during 3-48 h in the treatment group were significantly lower than those in the model group.
