乙醇预处理对肝脏缺血再灌流损伤的保护作用

摘　　要：目的　探讨胃饲乙醇预处理对大鼠肝脏缺血再灌注损伤的影响。方法　雄性Wistar大鼠随机分组 ,动物乙醇预处理后制作缺血再灌注模型 ,于术后 0到 72h内不同时相活杀大鼠 ,留取血及肝脏标本保存待检。结果　实验组和实验对照组较正常组ALT值明显升高 ,72h后基本恢复 ,实验组恢复较快 ,但实验组和对照间统计学差异不明显 ;病理观察见实验组较对照组表现为较轻的病理损害 ;免疫组化结果 :正常肝脏仅见弱阳性 ,散在分布于胞浆 ;术后随再灌注时相的延长 ,HSP70表达逐渐增强 ,实验组表达明显强于对照组和正常组 (P <0 .0 5 ) ;WesternBlotting显示HSP70阳性条带于再灌注 3h开始增强 ,6h明显增强 ,持续至 72h达到最强 ,实验组和对照组相差显著。结论　胃饲乙醇预处理能够明显增强缺血再灌注大鼠肝脏HSP70的表达 。Objective To investigate the expression of HSP70 in rat liver preconditioned by ethanol and its influences on the liver ischemia-reperfusion injury. Methods Adult male Wistar rats were divided randomly into two groups. Alcohol were given though gastric gavage 24 hours before 30-minute ischemia in rat liver in the experimental group. Rats were killed at different time points during the period of reperfusion. Blood and liver tissue samples were taken for examinaton. Results (1) There were obvious differences in pathological observation between experimental group and control group. Despite of a certain degree of inflammatory cell infiltration, there was no obvious necrosis in experimental group which was not like in the control group. HSP70 was found in normal liver cell plasma at a low level and increased after I/R injury by immunochemistry staining. HSP70 was significantly elevated after the ischemia-reperfusion injury in experiment group when compared with the control group (P<0.05). Western Blotting exmination showed that the hsp70 was enhanced at 3 hours after liver reperfusion and increased obviously 6 hours which lasted until 72 hours. There was obvious difference between two groups. Conclusions Ethanol pretreatment can induce the expression of HSP70 in rat liver and protect the liver from ischemia reperfusion injury.
