摘  要:目的 探讨下丘脑室旁核 (PVH)是否通过直接的神经联系对淋巴结的功能进行调控。方法 采用假狂犬病毒 (PRV)跨神经元追踪方法研究肠系膜淋巴结注射PRV大鼠PVH内PRV感染神经元的分布 ,同时采用免疫组化方法研究腹腔注射细菌脂多糖 (LPS)后大鼠PVHc fos表达的变化。结果 在注射PRV动物的PVH内出现被PRV标记的阳性神经元 ,既有大细胞神经元 ,也有小细胞神经元 ,它们主要分布在PVH尾侧帽部和内侧部。FOS阳性产物在PVH的神经元核中出现 ,其阳性细胞的分布范围及阳性细胞的数目在LPS注射组均明显大于对照组。两批不同实验动物的相同冠状平面比较 ,可发现PVH中大多数PRV标记的阳性神经元出现在LPS注射后FOS阳性细胞数目增多的区域内。结论 PVH可能通过直接的神经环路与淋巴结相互联系 。Objective To study the role of the neural pathway which connects the paraventricular nucleus of hypothalamus (PVH)and the lymph nodes of the immune system. Methods Pseudorabies virus(PRV,Batha strain)transneuronal tracing technique was employed to investigate the distribution of the neurons controlling the mesenteric lymph nodes in the PVH of the rat, and immunocytochemical methods was used to observe the changes of the c-fos expression in the PVH of rat after intraperitoneally injected bacterial lipopolysaccharide (LPS). Re- sults The PRV-labeled neurons including parvocellular and magnocellular neurons occured in the PVH at 96h following PRV injection. They were mainly localized in the dorsal cap region and the ventromedial parts of the caudal PVN. FOS protein expression appeared in neuronal nuclei within the PVH . Marked increases in LPS -induced FOS protein expression were observed. Comparing PRV injection rats with LPS injection rats in the equivalent coronary levels in the PVH, it was shown that most of PRV-labeled neurons distributed in the areas where the numbers of FOS positive neurons were increased after LPS immunization. Conclusions The results suggest that interactions between the PVH and the lymph nodes are mediated via the direct neural pathway communication and that PVH may be the integrative center of neuroimmunomodulation.
