氯沙坦对自发性高血压大鼠血浆sICAM-1水平和PAI-1活性的影响

摘  要:目的:探讨早期雄性自发性高血压大鼠(SHR)血浆可溶性细胞间粘附分子-1(sICAM-1)水平和纤溶酶原激活物抑制剂-1(PAI-1)活性的变化及氯沙坦对其的影响。方法:将20只雄性SHR随机分成2组,每组10只。其中1组灌喂氯沙坦20mg/kg·d,另一对照组给正常饮水,共12周,并与雌性同周龄Wistar Kyoto大鼠(WKY)10只比较。采用酶联免疫吸附法(ELISA)、发色底物显色法分别对3组大鼠血浆sICAM-1水平和PAI-1活性进行检测。结果:与WKY大鼠比较,SHR大鼠血浆sICAM-1水平和PAI-1活性明显增高(P<0.01);经氯沙坦治疗12周后SHR 血压显著下降,血浆sICAM-1水平和PAI-1活性亦明显降低。结论:早期雄性SHR大鼠血浆sICAM-1水平和PAI-1活性校WKY大鼠明显升高,氯沙坦在有效降压的同时能降低SHR大鼠血sICAM-1浓度和PAI-1活性。Objective: To investigate the level of plasma soluble intercellular adhesion molecule-1 (sICAM -1), activity of plasminogen activitor inhibitor -1 (PA1-1) and the effects of Losartan in early spontaneously hypertensive rats (SHR). Methods: Twenty SHR were randomly divided into 2 groups of 10 rats each. One group were fed with Losartan of 20mg/kg·d per day for 12 weeks, while the remained group (SHR control group) and 10 Wistar kyoto rats (WKY) were given normal water only for equal time. Enzyme linked immunosorbent assay (EL1SA), speclrophollnclric assay were used to measure the plasma level of sICAM -1. activity of PAI-1 before and after Losartan treatment. Results: It was shown that sICAM-1 level and PAI -1 activity of SHR control group were higher than WKY group (P<0. 01). After treatment of Losartan. the blood pressure (BP) in SHR significantly decreased (P<0. 01). Compared with the control, the sICAM-1 and PAI-1 decreased in treatment group of SHR (P<0. 01 ). Conclusion: Plasma level of sICAM-1 and activity of PAI-1 are elevated in SHR. Losartan not only decreased the blood pressure of SHR. but also decreased sICAM-1 level and PAI-1 activity.
