错位双链寡核苷酸中碱基比例的测定研究

摘　　要：使用碱和碱性磷酸酶降解错位双链寡核苷酸 (Poly :Ⅰ :C12 U) ,使其生成三种核苷 ,再使用反相 高效液相色谱 (RP HPLC)方法 ,分析核苷的组成 ,从而计算出PolyⅠC12 U中的碱基组成比例 ,结果表明样品中的碱基比例接近设计比例 1 2∶1。In this research, the reverse phase high preformance liquid chromatography method was used to analysis the base ratio in synthetic double stranded oligouncleotides (Poly Ⅰ:C 12U).After the sample was treated by alkali and alkaline phosphorylase, the result show that the rate of cytidine monophosphate to uridine monophospate in Poly Ⅰ:C 12U was near to designed ratio(12∶1). This method can be used in quality administration.;
