摘  要:目的 探讨对凝血酶 (Thro)、肾上腺素 (Adre)和腺苷二磷酸 (ADP)诱发的血小板聚集的影响 ,观察灯盏花素 (Bre)对血小板内游离钙 ( [Ca2 +]i)的作用。方法 ①利用血小板聚集仪 ,记录加入Bre前后Thro、Adre、ADP对血小板聚集的作用。②利用钙荧光指示剂Fura 2 /AM ,观察Bre对Thro、ADP引起的血小板内游离钙 ( [Ca2 +] i )的影响。结果 ①Bre10 -4mol·L 1对ADP诱发血小板聚集的抑制率为 18.4± 9% ,但对Thro和Adre诱发的血小板聚集作用无效。②Bre还可抑制ADP诱发的血小板内 [Ca2 +]i的升高 ,但对Thro诱发的血小板内 [Ca2 +]i 的升高无抑制作用。结论 Bre可抑制ADP诱发的血小板聚集 ,其机制可能与其抑制ADP升高血小板内 [Ca2 +]i 的途径有关。Objective To study the effects of Breviscapine(Bre) on platelet aggregation induced by Thrombin(Thro), Adrenaline (Adre),and ADP; and investigate the effects of Bre on intra platelet free calcium elevation induced by Thro and ADP. Methods ①Using platelet aggregation instrument, to record platelet aggregation curves induced by Thro, Adre, ADP before and after adding Bre.②Using fluorescence indicator_Fura 2/AM,to determine the effects of Bre on intra platelet free calcium elevation induced by Thro and ADP. Results ①Bre10 -4 mol·L 1 could inhibit platelet aggregation induced by ADP(the inhibition rate was 18.4±9%), but could not inhibit that induced by Thro and Adre.②Bre could also inhibit intra platelet free calcium elevation induced by ADP, but could not inhibit that induced by Thro. Conclusions The mechanism by which Bre inhibit platelet aggregation induced by ADP may be relate to its inhibiting effect on ADP induced intra platelet free calcium elevation.
