摘  要:目的 探讨丹参酮ⅡA (TanⅡA )诱导NB4细胞分化和凋亡过程中PML RARα融合蛋白变化及其与细胞分化、凋亡的关系。方法 采用免疫荧光法检测PML RARα融合蛋白的变化 ;同时计算细胞生长抑制率 ,观察细胞形态学变化 ,进行NBT还原实验 ,采用流式细胞术检测细胞凋亡和分析细胞周期。结果 TanⅡA能减少NB4细胞中微小荧光颗粒数 ,恢复NBs大斑点荧光颗粒 ,作用 72h后颗粒数减少最明显 ;同时 ,TanⅡA能提高细胞生长抑制率、诱导分化 ,增强NBT还原能力和提高凋亡细胞比率 ,降低细胞增殖指数 ,作用 12 0h后细胞分化和凋亡作用最强。结论 PMLObjective To investigate the relationship between the PML-RARαfused protein and the differentiation and apoptosis of NB 4 cells induced by TanⅡA.Methods The PML-RARα fused protein in NB 4 cells induced by TanⅡA was measured by immuno-fluorescence.The proliferative inhibition rate was calculated and the cell differentiation was demonstrated by morphology and NBT reduction assay.Cell apoptosis and cell cycle were measured by flow cytometry(FCM).Results TanⅡA reduced the number of green micro-punctuates and increased the number of big green NBs speckles in NB 4 cells,reaching the peak 72 hours later.TanⅡA elevated the proliferation inhibition rate,differentiation rate,NBT reduction ability and apoptosis rate,and decreased the proliferation index,especially 120 hours later.Conclusion The attenuation of PML-RARα fused protein is the mechanism of differentiation and apoptosis in NB 4 cells induced by TanⅡA.
