摘  要:目的 研究白藜芦醇 (RES)对受照射小鼠的辐射防护作用及其分子作用机理。方法采用6 0 Coγ射线照射小鼠模型 ,观察小鼠 30d存活和死亡动物存活时间。用原位末端标记法观察受照射小鼠脾细胞凋亡 ,流式细胞仪检测脾细胞凋亡率和凋亡相关蛋白的表达水平 ,同时检测脾细胞内凋亡相关酶活性大小。结果 照射前给予RES能明显提高受照射小鼠的存活率 ,延长死亡动物存活时间。RES能明显抑制受照射小鼠脾细胞凋亡 ,提高Bcl 2表达水平 ,对于Fas表达无明显影响 ,同时脾细胞内的Caspase 3和Caspase 8活性明显升高。结论 RES具有明显的辐射防护作用 ,其作用机理与抑制辐射敏感细胞的细胞凋亡有关。Objective To investigate radiation-protective effect of resveratrol and its molecular mechanism. Methods Kunming mice were ig administered with resveratrol before 60 Co γ-irradiation.Thirty-day survival rate and the average life span of dead mice post-irradiation were observed.The apoptosis of spleen from irradiatied mice was detected by FACS and in situ termianl labeling method.The effect of resveratrol on the activities of Caspase-3 and Caspase-8,and the expression levels of Bcl-2 and Fas were examined. Results Administration with resveratrol resulted in increases of 30-day survival rate and prolongation of average life span of the dead mice.Aoptotic rate of spleen cells decreased,expression level of bcl-2 increased,the expression of Fas did not change,and the activities of Caspase-3 and Caspase-8 increased in spleen cells of irradiation groups. Conclusion The results indicate that resveratrol has radiation-protective effect and its mechanism might be related with its suppression of apoptosis of radiation-sensitive cells. ;
