摘 要:目的 测定刺参酸性粘多糖经硫酸水解后葡萄糖醛酸的含量 ,以此作为刺参酸性粘多糖质控标准 ,考察刺参酸性粘多糖的稳定性。方法 样品采用硫酸水解咔唑比色法测定葡萄糖醛酸的含量。结果 最大吸收波长为 5 2 0nm ,葡萄糖醛酸的回归方程为 :Y =0 .0 14 2 3 +0 .0 1662X ;r =0 .9991,其浓度在 10~ 5 0 μg ml范围内线性关系良好。三批样品含量为 12 .91%、11.5 3 %、13 .3 7%,平均加样回收率为 99.49%,RSD =1.0 9%(n =9)。结论 本法简便易行 ,灵敏度高 ,重现性好 ,可用于刺参酸性粘多糖的质量控制 。Objective To determine the content of glucuronic acid after acid mucopolysaccharide in stichopus japonicus sekenka (SJAMP) was hydrolyzed by sulfuric acid and then took it as the standard to control the quality of stichopus japonicus sekenka acid mucopolysaccharide;to investigate the stability of SJAMP.Methods The content of glucoronic acid was tested by sulfuric acid carbazole colorimetry method.Results The maxium absorption was 520 nm and the linear equation regression was Y=0.01423+0.01662X,r=0.9991,with the range of 10~50 μg/ml,good linear relationship between absorption and concentration was found.By assaying 3 samples,the contents of acid mucopolysaccharide were 12.59%, 11.71% and 13.50% respectively;the average recovery ratio and RSD were 99.49% and 1.09% (n=9).Conclusion The method is simple,steady,accurate and reproducible,which can be used for the quality control of preparing stichopus japonicus sekenka acid mucopolysaccharide,and its appearance and contents are steady.
