番茄斑萎病毒属(Tospovirus)病毒在云南的发生分布研究初报

摘　　要：应用电子显微镜观察形态学及细胞病理特征,间接酶联免疫吸附测定(I-ELISA)和双抗体夹心ELISA(DAS-ELISA),部分样品经番茄斑萎病毒(TSWV)核壳体蛋白基因(N-gene)的反转录聚合酶链式反应(RT-PCR)扩增,以及部分样品的生物学回接测定的方法,对采自云南不同地区20种寄主植物上204份疑似Tospovirus侵染的病株样品进行了检测。结果表明,共有20种寄主作物139份样品中含有Tospovirus分离物,从检出样品的分布看,云南全省均有分布,但在海拔2400m以上的地区其症状表现不明显。Tospovirus云南分离物在血清型、细胞病理特征以及寄主范围方面与国内外报道的同属病毒有共同的特征,同时也有较明显的差异,即云南存在有Tospovirus新的株系或种类。The occurrence and distribution of Tospovirus in Yunnan has been studied. 204 samples collected from 20 kinds of host plants in different regions with the symptoms caused by Tospovirus infection were used in this study. These samples were used to delect the morphology and cytopatho logical characterization by observation under electron microscopy, and by indirect enzyme linked immunosorbent assay (I-ELISA) , by double antibodies sandwich ELISA (DAS-ELISA) , Some samples were detected by reverse transcription polymerase chain reaction (RT-PCR) with primers of nucleocapside protein gene (N-gene) of Tomato spotted wilt virus (TSWV) . And some samples were detected by inoculating host plants to test the characterezitic symptoms. Tospovirus isolates had been detected from 139 samples of 20 kinds of host plants. It is found that Tospovirus distributed over the Yunnan province, but the symptom did not appearance above 2 400 m altitudes. There were common characteristics on the serological type, cytopothology and host range of the Tospovirus Yunnan isolates with other Tospovirus isolates in other places or countries. At the same time, Tospovirus isolates Yunnan had differences with other Tospovirus isolates in terms of that there would be new or different strains or species of Tospovirus in Yunnan.
