摘  要:目的 探讨地甘口服液对环磷酰胺损伤所致小鼠脾组织凋亡及凋亡相关基因bcl-2和bax mRNA表达的作用。方法 建立环磷酰胺损伤的小鼠模型(小鼠用环磷酰胺100mg/kg,腹腔注射1次/d,连续3d,造成模型),喂饲100%地甘口服液每次0.2mL/10g小鼠,每日2次。第10天用原位末端标记的TUNEL技术检测脾组织细胞凋亡和原位杂交法检测凋亡相关基因bcl-2和bax mRNA的表达。结果 地甘口服液组与对照组比较,脾脏的细胞凋亡率和bax mRNA表达显著减少(P<0.01),bcl-2 mRNA的表达显著增强(P<0.01)。结论 地甘口服液可能通过促进bcl-2和降低bax mRNA的表达,促进造血细胞的增生和提高机体免疫力。Objective To study the effect of Digan oral liquid (DGOL) on expression of apoptosis in the spleen tissue and some apoptosis-related genes mRNA with CTX lesion mice. Methods After the model of chemical lesion given CTX of 100mg/kg mouse by intraperitoneal injection was established, the animals were fed with 0.2mL/10g of 100% DGOL. Apoptosis was detected by terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling (TUNEL) insitu hybridiation. Results In the DGOL group, the apoptosic cells ration was significantly decreased (P<0.01) and bcl-2 mRNA expression was significantly increased (P<0.01) in spleen tissue than control group. Conclusion DGOL can restrain the apoptosis of spleen tissue, due to the enhanced bcl-2 and the decreased bax in mRNA expression with radiation lesion, thus prevent the blood cells from injuring and premote the immunity.
