摘 要:目的 :进一步探讨川芎嗪对缺氧 /再复氧心肌细胞内游离钙的作用。方法 :培养新生大鼠心肌细胞缺氧 /再复氧模型 ,以荧光探针Fluo 3/AM负载 ,通过激光扫描共聚焦显微镜观察损伤和川芎嗪组心肌细胞内荧光强度。结果 :损伤组细胞内钙荧光强度明显增强 ,川芎嗪各组细胞内钙荧光强度均有不同程度的减弱 ,尤以低、中剂量组明显 ,与损伤组比较有显著性差异 (P <0 .0 1)。结论 :心肌缺氧 /再复氧细胞内游离钙显著增加 ,而一定剂量的川芎嗪可降低细胞内游离钙 。Objective: To study the effects of Lignstrazineon on free Calcium in culture myocytes during hypoxia and reoxygenation. Methods: A model from cultured myocytes of neonatal Wistar rat and hypoxia/ reoxygenation injured was established. The myocytes were loaded with Flou-3/AM, a Ca 2+ fluorescent indicator. The fluorescent intensitie of intercellular calcium concentration was measured by Laser Scanning Confocal Microscopy in each group. Results: Each treated group with Lignstrazineon could decreases intercellular calcium concentration especially in the lower and the middle dose groups,with had significantly difference compared with that in the injured group(P<0.01). Conclusion: The cultured myocytes could be effectively protected by Lignstrazineon in case of hypoxia/ reoxygenation through decreasing the intercellular calcium concentration.
