摘 要:目的 :观察牛珀至宝丹 (NPZBD)与鸡卵黄免疫球蛋白 (IgY)以及两药合用 (CNI)对内毒素休克大鼠肝细胞凋亡及形态学的影响 ,并探讨其机理。方法 :采用外源性内毒素休克模型 ,分组观测BPZBD、IgY、CNI对内毒素休克大鼠的平均动脉压 (MAP)、肝组织细胞凋亡和电镜下形态学的变化。结果 :除假手术组外 ,其余各组血压都明显下降 ,而牛珀组、免鸡组、牛免组的血压下降幅度明显低于其他组 ;模型组、普鸡组肝细胞凋亡显著高于对照组 ,而免鸡组、牛珀组、牛免组较模型组均显著减轻 ,其凋亡程度接近对照组 ;肝组织电镜结果显示对照组肝细胞核及线粒体形态正常 ,模型组的损伤最严重 ,普鸡组次之 ,而牛珀组、免鸡组、牛免组损伤较轻。Objective To investigate the effects of Niu Po Zhi Bao Dan(NPZBD)medicinal berbs and IgY on the apoptosis of the rats' livers and the morphology of the rats' liver cell with endotoxin shock and to approach the pathogenic mechanism.\;Methods Sixth Sprague-Dawley(SD)rast were randomized into six groups with 10 in each group.I NPZBD medicinal herbs and IgY treated group (CNI),Ⅱ NPZBD treated group,Ⅲ IgY treated group,Ⅳ shock-model group,V normal-control group,Ⅳ common-yolk treated group.Rats were all pretreated before experiment.In the normal-control group,the rats were injected physiologic saline,in the other groups,the rats were injected intravenously lethal dosage of E.coli Lipopolysaccharide(LPS).In two hours,observing the changes of Mean of E.coli Lipopolyasccharide(LPS).In two hours,observing the changes of Mean arterial blood pressure(MAP) of all groups by the BL-410 Biology Computer Systems.After two hours,assessed the apoptosis of liver with Flow cytometry(FCM);;histology for the cell of liver throug trasmission electron microscope(TEM).\;Results MAP of normal group was notably different from the others,MAP were markedly decreased in the Ⅳ,Ⅵ groups than that in the Ⅰ,Ⅱ,Ⅲ groups.The number of the apoptosis of liver in the Ⅳ,Ⅵ groups were higher significantly than the Ⅴ group (P<0.01).Compared with the Ⅳ group,the number of apoptosis in the Ⅰ,Ⅱ,Ⅲ groups,reduced notably(P<0.01) and was close to the normal level.the damage of mitochondrion and nucleus in the Ⅰ,Ⅱ,Ⅲ groups were more less than the Ⅳ,Ⅵ groups.\;Conclusions the results indicated that IgY?NPZBD?CNI all are able to resist rats with endotoxin shock by alleviating the damage of liver cell and decreasing the number of apoptosis of liver caused by endotoxin,and keep the degree of MAP of endotoxin shock rats.\;
