摘  要:本文从家兔肋软骨的生长板软骨分离了软骨细胞,并在离心管内进行高密度培养。这些软骨细胞在自身合成的基质中增殖、分化形成软骨组织。在组织学上,细胞的排列呈现出明显的规律性,细胞纵向排列成柱状并且肥大细胞位于上侧、增殖细胞位于下面形成明显的细胞分化区带即(增殖、成熟区和肥大区,)表现出与骺板软骨相类似的特点。当培养达25天时,成熟软骨细胞趋向于肥大化表现出全层肥大化的特点,同时还观察到在细胞外基质出现明显的钙化现象。Chondrocytes which isolated from the rabbit costal growth-plate cartilage were seeded at high density in centrifuge tube as packed mass and maintained in MEM medium supplemented with 10% fetal bovine serum, ascorbic acid and antibiotics. The chondrocytes started to proliferate and synthesized an abundant extracellular matrix 2 days after cell seeding. These chondrocytes reorganized into a growth-plate cartilage-like tissue by proliferating and differentiating in matrix synthesized by themselves. The chondrocyte cultured was organized longitudinally into cellular column and horizontally into cellular zones containing proliferating, maturing and hypertrophic cells. Finally, chondrocytes undergo terminal differentiation that synthesized an abundant alkaline phosphatase and extracellular matrix mineralization was revealed by alizarin red staining.
