摘  要:目的:鉴定沙美特罗在小鼠尿中的主要代谢产物。方法:ig给药后,收集小鼠尿液,经固相提取,葡萄糖醛酸酶水解,进行LC/MS/MS分析和硅烷化后进行GC/MS分析同时分离鉴定沙美特罗代谢产物。结果和结论:在给药后尿样中发现沙美特罗原型和4种代谢产物M1~M4,其结构推测为19-羟基沙美特罗(M1)、2-羰基沙美特罗(M2)、19-羰基沙美特罗(M3)和19-羟基-8-甲氧基沙美特罗(M4)。Objective:To study the metabolites of salmeterol, which has been forbidden by the Medical Commission of the International Olympic Committee(IOC) since 1995. Methods:Urine sam-ples of ten healthy mice were collected within 24h after ig administration of salmeterol. The metabolites of salmeterol(M1-M4) were identified by LC/MS/MS after solid phase extraction followed by enzyme hydrolysis and GC/MS after derivatization. Mass spectrometry was operated in the positive mode for the method of LC/MS/MS. Result and conclusion:Salmeterol and it's four metabolites (M1-M4) were identified both by LC/MS/MS and GC/MS. M1-M4 were deduced as 19-hydroxyl-salmeterol, 2-carbonyl-salmeterol, 19-carbonyl-salmeterol and 19-hydroxyl-8-methoxyl-salmeterol, respectively.
