摘 要:目的:观察大鼠全脑照射后早期主要炎性细胞因子含量的变化。方法:对SD大鼠用4MeV电子线单次全脑15 Gy和30 Gy照射后,用酶联免疫吸附试验法检测照射后6 h、1 d、1 w和1个月大鼠大脑皮层中IL-1β、IL-6和TNF-α的含量。结果:与正常对照组和麻醉后对照组相比,15 Gy单次全脑照射后1月内各组以及30 Gy后1 d、1 w组,大鼠大脑皮层上清波中三种细胞因子的含量无异常变化;在30 Gy照射后6 h组皮层中IL-1β和TNF-α有明显升高,含量达536 pg/g与339 pg/g,IL-6在大脑皮层中的含量没有差别。结论:大鼠全脑照射后6 h脑组织中就有一些炎性细胞因子的升高,它们在放射性脑损伤的发病机制中可能有重要的作用。Objective:To investigate the changes of some inflammatory cytokines at the early stage after brain irradiated in rats. Methods: The whole brain of SD rats was irradiated with the single dose of 15 or 30 Gy of 4 MeV electron beam. The enzyme-linked immunosorbent assay (FLISA) was used for the measurement of IL-1β, IL-6, and TNF-α content in the brain cortex tissue supernatant of rats at 6 hours,1 day,1 week and 1 month post-irradiation.Results:Compared with both the sham anaesthesia control group and the anaesthetized with chloral hydrate but sham irradiation group, there were no difference in levels of IL-1β, TNF-α and IL-6 among all the groups with 15 Gy irradiated rats, the 30 Gy post-irradiated 1 day and 1 week group. But the brain tissue supernatant levels of IL-1β and TNF-α at 6 hours post-irradiation with 30Gy group were significantly higher than that of control groups,which were up to 536 and 339 pg/g,respectively. While the level of IL-6 in brain of 30 Gy group was decreased in 1 month. Conclusion: The initial response of the brain to irradiation involves the expression changes of some inflammatory cytokines, which may play an important role in the pathogenesis of brain radiation injury.
