摘 要:目的 探讨中药方剂 (生牡蛎 30g,地龙 30g ,生黄芪 30g ,皂刺 9g,丹参 30g,川牛膝 9g,海藻 15g ,虎杖 2 0g,王不留行 15g ,当归尾 15g ,蒲公英 15g ,穿山甲 2 0g)对小鼠前列腺细胞增殖及凋亡的影响。 方法 体重 30~ 4 0g雄性昆明种小鼠 12 0只 ,随机分为 5组 :①正常对照组 ,②增生组 ,③阴性对照组 ,④中药方剂组 ,⑤非那雄胺 (保列治 )阳性对照组 ;每组 2 4只。正常对照组小鼠直接处死取前列腺观察湿重 ,对其余小鼠采用尿生殖窦植入方法建立小鼠良性前列腺增生 (BPH)模型 ,增生组小鼠于第 6 1天处死取前列腺观察湿重 ;其他组小鼠分别于第 6 1天开始喂生理盐水、中药方剂及保列治 ,于第 91天处死取前列腺观察湿重 ,应用流式细胞术 (FCM)检测 3组前列腺细胞增殖率及凋亡率的变化。 结果 增生组前列腺重量为 (14 9.30± 8.6 4 )mg ,中药方剂组为 (85 .6 0± 17.97)mg,两组相比差异有显著性意义 (P <0 .0 1)。增生组前列腺细胞凋亡率为 (3.6 6± 0 .73) % ,增殖率为 (2 6 .0 5± 3.0 2 ) % ;中药方剂组凋亡率为 (6 .6 7± 1.91) % ,增殖率为 (2 6 .2 8± 2 .84 ) % ;保列治组凋亡率为 (6 .2 6± 2 .4 2 ) % ,增殖率为 (2 6 .2 9± 4 .0 7) %。中药方剂组前列腺细胞凋亡率较增生组明显升高 (P <0 .Objective To investigate the therapeutic effects of traditional Chinese medicinal herbs(TCMH) on benign prostate hyperplasia(BPH) and their influence on proliferation and apoptosis of prostate cells in experimental mice. Methods A total of 120 male Kunming mice(weight,30 to 40 g)were randomly divided into 5 groups:①normal control group;②hyperplastic group;③normal saline(NS)(negative control)group;④TCMH group;⑤finasteride (positive control) group.Each group included 24 mice.The mice in normal control group were killed and their prostates were weighed.The BPH model was induced by implanting the urogenital sinus in the mice in other 4 groups.In the 61st day,the prostate weight of the mice in hyperplastic group was examined after they were killed;and the mice in the remaining groups were fed with NS,TCMH and finasteride,respectively.In the 91st day,the mice in these 3 groups were killed and their prostates were weighed.The proliferation and apoptosis of the prostate cells of mice in the 3 groups were determined by flow cytometry(FCM). Results The prostates in hyperplastic group [(149.30±8.46)mg] were heavier than those in TCMH group [(85.60±17.97)mg] ( P <0.01).Rates of apoptosis and proliferation in hyperplastic group were (3.66±0.73)% and (26.05±3.02)%;those in TCMH group were ( 6.67 ±1.91)% and (26.28±2.84)%;and those in finasteride group were (6.26±2.42)% and (26.29±4.07)%,respectively.The apoptosis rate in TCMH group increased significantly( P <0.01),while the proliferation rate changed very slightly( P >0.05). Conclusions The TCMH has a significant therapeutic effects on experimental mice with BPH.The mechanisms may be as follows:TCMH can regulate the relevant genes of the prostate cells so that they can promote the apoptosis of the prostate cells and as a result,the volume and weight of the prostate of the mice are reduced.
