摘 要:目的 探讨黄精多糖对糖尿病鼠心脏和肾脏组织糖基化终产物受体mRNA (RAGEmRNA)表达的调节作用。方法 腹腔注射链脲佐菌素建立动物模型 ,各组实验动物心、肾组织RAGEmRNA表达的测定采用RT PCR法。结果 糖尿病模型组实验鼠心、肾组织RAGEmRNA表达增加 ,其RAGE/β actin相对值明显高于对照组 (P <0 0 1)。应用黄精多糖治疗后 ,治疗组实验鼠心、肾组织RAGEmRNA表达较模型组降低 ,其RAGE/β actin相对值显著低于模型组 (P <0 0 5 )。 结论 黄精多糖能抑制糖尿病鼠心、肾组织RAGEmRNA表达 ,对高血糖及糖基化终产物 (AGE)造成的组织损伤具有保护作用。Objective To study the regulation function that polygona-polysaccharose to the expression of RAGE mRNA in diabetic rats heart and kidneys.Methods Diabetic rats model was made by injecting streptozotocin into its abdominal cavity. RT-PCR method was used to examine the expression of RAGE mRNA in rats' heart and kidneys.Results RAGE mRNA in heart and kidney tissue of diabetic rats was increased. The value of RAGE/β -actin in diabetic rats was significantly higher than that in the control group (P<0.01).After the treatment with polygona-polysaccharose,the expression of RAGE mRNA in tissue of heart and kidneys was decreased in the control group. The RAGE/β-actin value in the treated group was significantly lower than that in the animal model group (P<0.05).Conclusion Polygona-polysaccharose may decrease the expression of RAGE mRNA in the tissues of heart and kidney in diabetic rats and has protective effect on the tissue damage caused by hyperglycemia and AGE.
