摘 要:目的 观察莪术油明胶微球栓塞Beagle犬肝动脉后的病理改变 ,为临床实验提供依据 .方法 Beagle犬随机分为生理盐水组 (10ml/kg)、空白微球组 (15mg/kg)、莪术油微球低剂量组 (7.5mg/kg)和莪术油微球高剂量组(15mg/kg) ,每 4周经肝动脉给药 1次 ,连续给药 3次 ,停药后观察 4周 .观察肝病理组织学改变 .结果 明胶微球肝动脉栓塞可致肝组织不可逆性损伤 ,高剂量组广泛全小叶坏死 ,低剂量组灶性坏死 ,空白微球组改变类似高剂量组 ,生理盐水组未见肝组织损伤 .结论 莪术油微球重复经Beagle犬肝动脉给药 。Objective To observed The pathological change of Zedoary turmeric oil gelatin microspheres administered via hepatic artery on the beagle dog. Methods Eighteen Beagle dogs were randomly divided into experimental and control groups. Four groups included ZT-GMS high dosage group (ZT-GMS-H, 5 dogs) receiving ZT-GMS 15mg/kg every 4 weeks, ZT-GMS low dosage group (ZT-GMS-L, 5 dogs) receiving ZT-GMS 7.5mg/kg every 4 weeks, vehicle control group (4 dogs) receiving gelatin microspheres 15mg/kg every 4 weeks, blank control group (4 dogs) receiving physiological saline 10 mL/kg (identical with the volume required for microspheres injection) every 4 weeks. The dogs were administered likewise every 4 weeks, totally 3 times. We observed the liver pathological change. Results The tissue sample of ZT-GMS-H group and vehicle control group exhibited whole lobule coagulation necrosis in the hepatic tissue. The remaining hepatic tissue exhibited extensive moderate cellular edema with some cellular inflammatory edema in the necrotic region. The tissue sample of a deceased dog in the experiment exhibited whole lobule coagulation necrosis in the hepatic tissue. The tissue sample of two dogs sacrificed on day 30 after the last administration exhibited whole lobule coagulation necrosis in the hepatic tissue and mild cellular edema in the remaining hepatic tissue with no organization observed. The tissue sample of ZT-GMS-L group exhibited extensive spotty coagulation necrosis in the middle region with neutrophilic leukocyte infiltration. But the tissue sample of blank control group were no abnormal change. Conclusions The study demonstrated that ZT-GMS infused via hepatic artery may cause irreversible ischemic necrosis of the liver.
