薄层扫描法测定蚂蚁药材及其风湿克酒中亮氨酸的含量

摘　　要：目的 :建立蚂蚁药材及其风湿克酒中亮氨酸含量的测定方法 ,为控制蚂蚁药材及其制剂的质量提供依据。方法 :样品经薄层层析、2 %茚三酮乙醇溶液显色后 ,于λs=480nm、λR =650nm波长处扫描测定。结果 :样品的平均回收率为100 14 % ,RSD=3 4 %。暂定蚂蚁药材含亮氮酸不得少于1 0 % ,风湿克酒含亮氨酸每1ml不得少于0 44mg。结论 :本法可作为山西地方药材蚂蚁(黑蚂蚁 )及其制剂风湿克酒中亮氨酸含量的测定方法。OBJECTIVE:To establish a method for determination of the contents of leucine in ant medical material and its preparation-fengshike wine,so as to provide a basis for controlling the quality of ant medical material and its preparaˉtion.METHODS:After thin layer chromatography and colour-development with2%ninhydrin ethyl alcohol solution,the sample was scanned atλ S =480nm,λ R =650nm.RESULTS:The average recovery of the sample was100.14%,RSD=3.4%.The content of leucine in ant medical material was tentatively fixed at1.0%or more and that in Fengshike wine at0.44mg/ml or more.CONCLUSION:The method may be used for determination of the contents of leucine in Shanxi local medical material-ant(black ant,Formica fusca Laureile)and its preparation-Fengshike wine.
