银屑病角朊细胞及外周血单个核细胞RANTES的表达

摘　　要：目的　探讨 β 趋化因子在银屑病炎症细胞浸润中的作用。 方法　我们选择了 2 0例银屑病患者的皮损及其血清标本 ,以 5例正常人皮肤及血清做正常对照。应用免疫荧光及酶联免疫吸附测定方法分别检测表皮角朊细胞及血中单个核细胞RANTES的表达。结果　银屑病角朊细胞RANTES着染数为 (78 6 0±4 2 2 0 )个 ,血清及血中单个核细胞产生RANTES的量分别是 (1 2 0± 0 6 4 ) μg/L及 (10 12± 5 10 ) μg/L。与正常对照组相比 ,银屑病角朊细胞及外周血中表达水平均显著升高。结论　本研究结果可解释银屑病皮损中活化T细胞的迁徙 ,且肯定了RANTES在银屑病炎症过程中的作用。Objective To determine the role of β-chemokines in the migration of leukocytes in psoriasis.Methods We collected 20 skin biopsies and sera from patients with psoriasis and five biopsies and sera from healthy individuals as normal controls. RANTES (β-chemokine) was determined in the epidermal keratinocytes by immunofluorescence microscopy and in the peripheral blood mononuclear cells (PBMC) by ELISA respectively. Results The number of keratinocytes stained for RANTES in high power fields were 78.60±42.40 in psoriatic tissue. The serum RANTES levels and production of RANTES by PBMC were (1.20±0.64) ng/ml and (10.12±5.10) ng/ml respectively. The expression of RANTES in keratinocytes and PBMC of patients with psoriasis was significantly higher than that in normal control.Conclusion These findings provided an explanation for migration of activated T cells in psoriasis and substantiated the role of RANTES in the inflammatory process of psoriasis.
