摘 要:目的 观察黄芩苷、栀子苷对神经细胞缺氧缺糖/再灌注损伤的保护作用,在细胞水平阐明黄芩苷、栀子苷治疗脑缺血/再灌注损伤的作用机制。方法 选用SH-SY5Y细胞,采用缺氧、复氧结合缺糖、复糖的方法,对其进行攻击,模拟人体神经细胞的缺血再灌注损伤,观察黄芩苷、栀子苷对此损伤的治疗作用。结果 缺氧、缺糖8 h结合复氧、复糖12 h对神经细胞可造成明显损伤,导致SH-SY5Y细胞突起皱缩、变圆,可见有部分细胞漂起,细胞轮廓不清,凋亡率明显上升,线粒体的活性下降;而黄芩苷、栀子苷可以显著减轻上述损伤。结论 黄芩苷、栀子苷对缺氧缺糖/再灌注损伤的神经细胞有保护作用。OBJECTIVE: To elucidate the mechanism underlying the protective effects of Chinese herb baicalin and gardenoside on oxygen-glucose deprivation and reperfusion injury. METHODS: SH-SY5Y cell were subjected to the oxygen-glucose deprivation and reperfusion injury, and the effects of baicalin and gardenoside on neurocytes were examined. RESULTS: 8 hours of oxygen-glucose deprivation plus 12 hours of reperfusion led to severe injury to neurocytes. The activity of mitochondria decreased significantly, and the percentage of apoptotic cells increased significantly. It was also observed that baicalin and gardenoside could significantly alleviate this injury. CONCLUSION: Baicalin and gardenoside had direct protective effects on neurons subjected to oxygen-glucose deprivation and reperfusion injury, which might be one of the mechanisms underlying its therapeutic effects on oxygen-glucose deprivation and reperfusion injury.
