摘 要:目的 研究重组人组织型纤溶酶原激活剂改构体(reteplase)体外复性。方法 通过研究复性所需的时间、温度、pH值、蛋白浓度、氧化还原对比例和添加精氨酸等因素,确定了reteplase复性所需的基本条件,并对直接稀释法、氧化复性后再稀释和脉冲稀释法进行了方法学比较研究。结果 脉冲稀释法优于直接稀释法和氧化复性后再稀释。结论建立了reteplase复性方法,此方法可使重组蛋白在较高的重折叠浓度下仍具有良好的复性效率。OBJECTIVE: To study on the renaturation of reteplase in vitro. METHODS: The basic renaturation conditions of reteplase were established by analysis of time, temperature, pH, protein concentration, the ratio of reduced and oxidized thiol reagents, adding L-arginine, et al. The traditional dilution, oxidation renaturation before dilution and step-wise dilution were investigated. RESULTS: Compared with the traditional dilution and oxidation renaturation before dilution, the activity of reteplase obtained by step-wise dilution was significantly improved. CONCLUSION: The method of reteplase renaturation was established. The process above can be used to obtain reteplase with high concentration and high activity from Escherichia coli.
