摘  要:采用腹腔注射和雾化吸入卵蛋白建立小鼠过敏性哮喘动物模型 ,应用流式细胞术和免疫荧光法观察脾及肺泡灌洗液 (BALF)中T细胞数、T细胞周期和Bcl 2基因表达的变化 ,结果发现哮喘组脾和BALF中淋巴细胞CD3表达率显著高于正常对照组 (尤其是BALF明显 ) ;脾T细胞S期及G2 +M期细胞数明显增多 ,同时凋亡率增加 ;Bcl 2基因表达率显著增高。提示哮喘发病时 ,T细胞增多 ,脾T细胞活化增殖增加 ,凋亡增加。同时淋巴细胞BclTo investigate the changes of T lymphocyte count, T lymphocyte cell cycle and the expression of Bcl 2 in allergic asthma,20 male kunming mice of 5 weeks were randomly divided into control group and asthmatic group. An animal model of asthma was established by means of OVA sensitizing-challenging. CD3 expression in spleen and bronchoalveolar lavage fluid (BALF),T lymphocyte cell cycle and Bcl 2 expression in spleen were detected by flow cytometry. The Results showed that in BALF lymphocyte and in spleen lymphocyte, CD3 expression rate in asthmatic group was significantly higher than that in control group. In spleen lymphocyte, the cell count in S age, G2+M age and apoptosis rate weas significantly higher than that of control group, and the Bcl 2 expression rate was significantly higher than that of control group. In conclusion,in the allergic asthma mice model established in this experiment, T lymphocyte count increases significantly especially in BALF, meanwhile proliferation, activation of and apotosis rate of T lymphocyte increases in spleenic lymphocytes. Bcl 2 expression also increases.
