摘  要:目的:寻找一种适合于复方壳多糖人工皮肤的保存方法。方法:采用标本沉降式液氮冻存方法,将培养1周的人工皮肤冻存;并在冻存后1,2个月时解冻,继续器官型培养,观察种子细胞的生长情况,并通过组织学检查比较两者在细胞形态方面的差异。结果:解冻后的人工皮肤生长良好,组织学表现与没有冻存过的人工皮肤相比,未见明显不同。结论:液氮冻存没有改变复方壳多糖的生物学特性,提示该保存方法可能会在今后临床应用中发挥作用。AIM:To explore a suitable method for preservation of composite chitosan artificial skin. METHODS:The artificial skins were frozen with cryprotective agent(CPA) after 1 week culture by adopting sample sedimentation cryopreservation with liquid nitrogen.In the 1st and 2nd month after submersion in liquid nitrogen,the thawed artificial skins were conducted with organotypic culture.Viability of the seed cells were observed,and the morphological differences between the keratinocytes and fibroblasts were compared by histological observation. RESULTS:The thawed artificial skins were in a good status of growth.Histological examination showed that there were no differences between cryopreservation and nonpreservation skin at the forms of cells. CONCLUSION:Cryopreservation in liquid nitrogen with CPA has no obvious side effect in the biological characteristics of composite chitosan artificial skin,which indicates that this method for preservation can play an active role in further clinical application.
