摘  要:目的:测定给豚鼠肌注丹参注射液后血液、脑脊液和耳蜗外淋巴液中丹参素的含量。方法:采用高效液相色谱法(HPLC)中的内标法测定丹参素。色谱柱:ZORBA×Extend-C18(4.6mm×250mm,5μm)。流动相:甲醇-水-醋酸(22∶78∶0.25);流速:1.0mL/min;检测波长280nm;柱温:室温。结果:豚鼠肌注丹参注射液后可吸收入血液,80min时丹参素含量达最大值,为585.84mg/L。但并未进入脑脊液和耳蜗液。结论:此法可用于检测丹参注射液在豚鼠血浆中药代动力学特性及耳蜗组织中的靶向定位特征。丹参素可吸收入血液,但不能通过血脑屏障以及血-迷路屏障。AIM:To measure the content of danshensu in guinea pig blood,cerebrospinal fluid and perilymph fluid of cochlea after injection with danshen injection. METHODS:The high performance liquid chromatography(HPLC) was used for determination of danshensu by internal standard method.Chromato column was ZORBA×Extend C18(4.6 mm×250 mm,5 μm). Methanol water acetic acid(22∶78∶0.25) was used as mobile phase at 1.0 mL/min,the UV detector was set at 280 nm,and column temperature was room temperature. RESULTS:Danshensu could be absorbed into blood of guinea pig muscle along with danshen injetion.The concentration reached the highest level,585.84 mg/L,at 80 minutes,while danshensu could not be found in cerebrospinal fluid and perilymph fluid. CONCLUSION:The present method can be applied to detect the pharmacokinetic properties of danshan injection in blood plasm and target location characteristic of danshan injection in perilymph of cochlea tissue in guinea pig.Danshensu cannot pass through blood brain barrier and blood labyrinth barrier though it is aborbed into blood.
