摘 要:研究了酶反应的温度、pH值、底物浓度和反应时间等对kluyveromycesfragilisβ-D-半乳糖苷酶催化合成低聚半乳糖的影响.结果显示,以乳糖为底物,kluyveromycesfragilisβ-D-半乳糖苷酶催化合成低聚半乳糖的最适温度为60℃,最适pH值为6.0.酶法合成低聚半乳糖的产率随着底物浓度增加而增加.kluyveromycesfragilisβ-D-半乳糖苷酶催化低聚半乳糖合成的初速度较快,随着反应时间延长,反应速率降低,反应20h酶反应达到平衡.以660mg/mL乳糖为底物,在最适酶反应条件下,酶法合成低聚半乳糖的产率为207.12mg/mL.The aim of this research was to quantify the effect of temperature,pH,substrate concentration and reaction time on enzymatic synthesis of galacto-oligosaccharide.The optimal temperature and pH for galacto-oligosaccharide production by using β-D-galactosidase from kluyveromyces fragilis were 60 ℃ and 6.0,(respectively.)The higher galacto-oligosaccharide yeild was obtained at higher substrate concentration.Enzymatic reaction speed was fast at the beginning of reaction,then decreased,and chemical balance was reached after 20 hours incubation at 60 ℃.207.12 mg/mL of galacto-oligosaccharide was produced from 660 mg/mL lactose at optimal reaction conditions.
