肾综合征出血热病毒经消化道传播的实验研究

摘  要:将实验动物Wistar大鼠分为四组,用HFRS病毒AR90162株感染。A组用毒株悬液灌胃,B组让其自然食入含病毒的毒饵,C组经破皮感染,D组为“空白”对照。结果,A、B、C 3组均于感染后10d从尾血中查到特异性抗体,3组抗体累积阳性率分别为4/7、4/9.5、4/9.5;肺脏特异性抗原检出率3组分别为2/6,2/9,2/9。脏器抗原定位以肺为最多,其次为肝、脑,胃中没有查到抗原,肠道中仅A组1例;各组粪尿抗原均于感染后10~15d查到,但阳性率最高只有2/10。我们还对Wistar大鼠胃排空时间及胃内容物平均pH值进行了检测,结果胃排空时间平均为6h,胃内容物平均pH值为3.2±2.1。综合分析实验结果,我们认为消化道传播是可以实现的,但有条件性,如毒株的毒力、胃内容物pH值、饮食因素、外环境条件等均可影响经此途径的传播。Wistar Rat was used as the experimental animal, and HFRS Virus,AR90162 strain as infecting virus. The animals were divided into four group, named A,B,C,D. Group A was fed virus suspension with a special pipe; Group B took food with virus suspension; Group C was infected via skin lesion, Group D taking general food was designed as empty control. Antibody against HFRSV in sera of the animals was detected with IFA and the antigen in urine and stool with ELISA, once every 5 days after infection. All the animals were scarifised on day 35 to investigate the antigen in the organs such as lung, kidney, brain, liver with direct FA. The tissues showing positive result were used to isolate virus. The results were: specific IgG antibody could be detected from sera taken 10 days after infection and antigen in urine and stool taken 10~15 days after infection. The positive rate of antibody for group A, B, C were 4/7,4/9,4/9.5, respectively, and 2/7,2/9.5,2/9.5 for the antigen in urine and stool. There was no remarkable difference among the 3 groups,The distribution of viral antigen in the organ was detected in the following order: Lung>kidney>brain>liver>investine. Meanwhile, viruses isolated were obtained from lungs of A_6, A_(10), B_7, C_5, The average time of stomach empting, for wistar rat was about 6 hears, and the average value of pH for the content of stomach is about 3.5. the result suggested that the transmission of HFRS virus through digestive tract could be realized, but in general this mode was not easy.
