摘  要:目的 :研究一氧化氮合酶 (nitricoxidesynthase,NOS)抑制剂Nω 硝基 L 精氨酸甲酯 (Nω nitro L argininemethylester,L NAME)对急性一氧化碳 (carbonmonoxide,CO)中毒大鼠血脑屏障 (blood brainbarrier,BBB)通透性的影响 ,探讨NO介导CO中毒后BBB损伤的作用 .方法 :建立实验性CO中毒大鼠模型 .将健康雌性SD大鼠 4 0只随机分成4组 (对照组 ,CO中毒组 ,染毒前 1 5min给L NAME组和染毒后 30min给L NAME组 ) ,每组 1 0只 .用分光光度计法检测各组大鼠脑组织内伊文思蓝 (Evansblue ,EB)的含量 ,进行比较 .结果 :CO中毒后 2 4h ,大鼠大脑BBB通透性增加 ,脑组织内EB含量 (2 7.6± 1 .2 ) μg/g与对照组 (6 .1±1 .9) μg/g比较明显增高 (P <0 .0 1 ) ;给予L NAME治疗的两组大鼠大脑BBB损伤减轻 ,脑组织内EB含量分别为 (1 4 .8± 2 .3) μg/g和 (1 6 .0± 2 .2 ) μg/g ,较CO组 (2 7.6± 1 .2 ) μg/ g明显降低(P <0 .0 1 ) .结论 :急性CO中毒可导致大鼠大脑BBB通透性增高 ,中毒早期应用NOS抑制剂L NAME能够明显减轻大鼠大脑BBB损伤 。AIM: To determine whether NO mediates blood brain barrier (BBB) injury after acute CO poisoning in rats by investigating the effects of an NO synthase (NOS) inhibitor, Nω nitro L arginine methyl ester (L NAME), on BBB in CO poisoned rats, and to explore the possible mechanisms involved in this process. METHODS: The experimental rat models of CO poisoning were established. Forty healthy female Sprague Dawley rats were separated into four groups. L NAME (30 mg/kg, introperitoneally) was injected 15 min before or 30 min after CO exposure. Changes in BBB permeability were determined by detection of Evans blue (EB) content in rat brains with spectrophotometer 24 h after CO exposure. RESULTS: Compared with that in control group[(6.1±1.9) μg/g], a significant increase in EB content was found in the rat brains that were CO poisoned but not treated with L NAME[(27.6±1.2) μg/g] ( P <0.01). However, EB extravasation was significantly restricted in groups that were treated with L NAME 15 min before[(14.8±2.3) μg/g ]or 30 min after CO exposure [(16.0±2.2) μg/g ]( P <0.01). CONCLUSION: The BBB permeability increases after acute CO poisoning, which can be significantly restricted by pretreatment with NOS inhibitor, indicating that BBB injury caused by acute CO poisoning is partially mediated by NO.
