髓清丸对黑色素瘤VEGF MMP-2基因表达的影响

摘　　要：在恶性肿瘤的生长和转移中 ,肿瘤血管的生成起着关键作用 ,研究显示髓清丸可以抑制恶性肿瘤的转移 ,并对肿瘤血管生成有影响 ,可以抑制VEGF、MMP -2蛋白的表达 ,但尚不知其是否对VEGF、MMP -2mRNA表达有影响。目的 :研究中药活血化瘀复方髓清丸是否对肿瘤血管生成相关VEGF、MMP-2基因表达有影响。方法 :采用BALB/C无胸腺裸小鼠右侧腑窝皮下接种造成黑色素瘤B16自发转移实验模型 ,用RT -PCR技术检测了裸鼠腋下B16黑色素瘤组织中VEGF、MMP -2mRNA表达的改变。结果 :髓清丸高、低剂量组的VEGF、MMP -2基因的表达均较对照组下降。结论 :髓清丸能抑制肿瘤细胞的VEGF、MMP -2基因的表达。实验发现 ,对照组的癌细胞VEGF、MMP -2基因的表达较强 ,髓清丸高、低剂量组VEGF、MMP -2基因的表达均下降 ,这表明髓清丸似能抑制肿瘤细胞的VEGF、MMP -2的表达。可以认为 ,髓清丸有可能通过下调VEGF、MMP -2基因的表达 ,进而抑制VEGF、MMP -2蛋白表达 ,抑制肿瘤血管的生成 。Objective:The objective is to study the effect of Sui-Qing-Wan(SQW) on The Gene Related with angiogensis in B16 melanoma. Methods:B16 melanoma cell line, B16 cells, were injection into the right armpits of BALB/C nude mice to produce a model lung metastasis. Mice were randomly divided into three proups as did above. Animals were killed after 21 days. The gene expression of VEGF and MMP-2 were determined by reverse transcription polymerase chain reaction (RT-PCR) method. Results:The gene expression of VEGF in tumors was markedly inhibited by two dose of SQW compared to the control (P<0.01,P<0.01). The gene expression of MMP-2 in tumors was also markedly inhibited by two dose of SQW compared to the control(P<0.01,p<0.01). Conclusion:The gene expression of VEGF, MMP-2 were markedly inhibited by SQW.
