摘  要:目的 对皮层神经干细胞向胆碱能神经元定向分化及骨架蛋白 (β tubulin)进行探讨研究。方法 采用细胞培养技术、免疫细胞化学方法观察皮层神经干细胞向胆碱能神经元定向分化及其过程中骨架蛋白表达变化。结果 皮层神经干细胞在去除丝裂原后开始进行分化 ,至第 7d可分化为成熟神经元 ,NSE、AchE反应明显阳性 ,随着分化神经元的逐渐成熟 ,β tublin在细胞内的分布亦发生变化 ,由核周集中分布变化为广泛分布于细胞质内 ,形成细网状 ,构成细胞骨架。结论 皮层神经干细胞定向分化为胆碱能神经元的过程中伴随有骨架蛋白的时空变化 ,随神经元的成熟而构成细胞骨架。Objective To observe the directional differentiation of cortex neural stem cells into cholinergic neurons, and the changes ofβ-tubulin in this process. Method The differentiation of neural stem cells into mature cholinergic neurons was observed by cell culture technique and immunocytochemistry. Result The stem cells isolated from the cerebral cortex were induced to differentiate after the removal of mitogen. Mature neurons with positive NSE and AChE reaction were not observed until the 7 thday. With the maturation of the neurons, the distribution ofβ-tubulin in the cells was also changed from the perikaryal pattern, and the cytoskeleton (internal framework of a cell) was formed. Conclusion The distribution of β-tublin is changed in the directional differentiation of cortex neural stem cells into cholinergic neurons.
